In Vtiro Anti-Influenza Virus Activities of Polysaccharides from Antrodia cinnamomea in Submerged Culture

  Anti-influenza virus activities of polysaccharides extracted from Antrodia cinnamomea were carried out in Madin-Darby canine kidney (MDCK) cells and in chicken eggs.The growth of influenza H5 virus was obviously inhibited by Antrodia cinnamomea extracts (ACE) in MDCK cells when the cells were incubated with H5 Virus for 2 hours.The results indicated that 50% cytotoxic concentration (CC50) of Antrodia cinnamomea extracts (ACE) was 27.344±9.726 mg/mL in MDCK cells,and 50% maximal effective concentration (EC50) was 0.095±0.041 mg/mL.The selectivity index (SI,CC5o/ EC5o) was then calculated as 288.Furthermore,anti-influenza virus activities of ACE were tested in specific pathogen free embryonated eggs (SPF eggs) which were infected by H9 virus for 120 hours.The results indicated that the infected SPF eggs were well alive when they were treated by ACE at the dosage of 10 and 20 mg/egg,respectively.Compared with the control group,the activity of H9 virus was not significantly inhibited at the dosages of ACE of 5 and 10 mg/egg.But the viral inhibition ratio was observed up to 18% at the dosage of 20 mg/egg.In conclusion,current data suggested that ACE could have anti-influenza virus activities by inhibiting the growth of influenza H5 virus in MDCK cells and by reducing the amount of virus in H9-infected SPF eggs.
