The MatA Mating Type Locus of Pleurotus ostreatus

  The edible mushroom Pleurotus ostreatus is one of the most cultivated species worldwide.Fruiting bodies emerge on the dikaryon and this requires well-coordinated actions of the mating type genes.Pleurotus species are characterized by a tetrapolar mating incompatibility system which bases on two unlinked mating loci matA and matB with multiple alleles each.Every monokaryon has a specific mating type defined by its individual matA and matB alleles.Two monokaryons are compatible only in case of different alleles at both mating type loci.Only compatible monokaryons form a fertile dikaryon.In current research,we are elucidating the structure of different alleles of the matA locus.The locus contains genes for two distinct types of homeodomain transcription factors.We are cloning the genes from strains of different A mating type activity.In the future,we want to transform the genes into suitable monokaryotic hosts for functional analyses.As a first morphological marker,we will observe clamp cell formation at hyphal septa of resulting transformants.To study effects of transformed matA genes on later steps of development such as initiation of mushroom production and meiotic basidiospore formation,transformants might be appointed in classical genetic crosses and progeny analyses.To this end,reliable conditions for fruiting of dikaryotic strains in the laboratory are required.We therefore describe here also successful experiments of mushroom production under laboratory conditions.
