Functional Importance of Ribosomal Protein L4 (RPL4) Implicated In EBNA1-mediated Transactivation

  EBNAl,a nuclear antigen encoded by Epstein-Barr virus (EBV),was previously reported to be involved in the viral episome maintenance and DNA replication in EBV latent infections.Although EBNA l lacks any enzymatic activities,cellular proteins were proposed to be associated with EBNAl to carry out its transcriptional activation from episome.It was found previously (Dr.Peng et al,unpublished data,Tzu-Chi University) that 60s ribosomal protein L4 (RPL4) was associated with EBNAl and identified by pull down assay,followed by LC-MS-MS analyses.RPL4 resides in the 28S rRNA of 60S subunit in eukaryotes,while is located in the 23S rRNA of 50S subunit in prokaryotes.RPL4 appears to play an important role in early assembly of ribosome because RPL4 activates and stabilizes some ribosomal proteins in the 60S subunit.It was reported that RPL4 is crucial in both transcription and translation.In this study,we seek to uncover the molecular importance of RPL4 involved in EBNAl-mediated transcriptional activation following interaction of EBNAl with RPL4.OriP-Luc reporter system was constructed previously,and a series of deletion mutations of RPL4 were performed.Deletion mutation analysis of RPL4 showed aal-238 is essential in facilitating the EBNAl-mediated transcriptional activation,whereas aa241-287 is not required.These data suggested that Guα (RNA hlicase Ⅱ),proposed binding to aa254-333 of RPLA,may not be associated with the function of RPL4.The scientific knowledge gained from this study therefore contributes to the elucidation of functional roles of RPL4 involved in EBNAl-mediated transactivation and potentially important for the development of anti-EBV therapeutic agents.
