Microcalorimetric characterization of a marine bioflocculant-producing bacterium Halomonas sp.V3a

A microcalorimeter was used to measure metabolic activity by analyzing the thermodynamic parameters (growth rate constant κ,total heat released QΥ and heat released during the log phase QLog)of Halomonas sp.V3a growth under different conditions, that was found to yield a bioflocculant with high flocculating activity for Kaolin suspension. The results show that strain V3a had the most metabolic activity for glucose, the lowest for maltose with fixed nitrogen source (NH4Cl).The metabolic activity of strain V3a did not depend on the nitrogen content of selected compounds.Biomass concentration and κ value exhibited the similar change trend, but not consistent with QLog.Another phenomenon could also be found that flocculating activity is higher, biomass concentration or k is lower contrarily, for example, tryptone as carbon source. So,it elucidated that higher biomass can not represent the higher production of bioflocculant.
