Research of fermentation, separation and purification in production of extracellular glucan from Sclerotium rolfsii

  The glucan from Sclerotium spp.fermentation process can be used as oil-displacing agent applied in oil production.It is defined that by single factor test and orthogonal test the optimal conditions were:glucose 5%,sodium nitrate 0.4%,initial pH 5.0 and rotation speed 140 r/min.The rotation speed was the factor influenced the glucan production mostly in all factors.Raising rotation speed,the polysaccharides of mycelium and extracellular was increased in fermentation broth.The yield of crude polysaccharide get by extracting of alkali method was higher than that by extracting of water method.The polysaccharide get by water extraction was more suitable to be used as oil-displacing agent.The deproteinization effects of enzymolysis and ultrafiltration were more outstanding than that of Sevag method.
