Evaluation of the Synergistic Mutagenicity between Hexavalent Chromium and 1-(4-amino-2-methyl-5-pyrimidinyl) Methyl-3-(2-chloro-ethyl)-3-nitrosourea at Low Levels by Ames Test

  Chromium is a widely distributed and very persistent anthropogenic heavy metal pollutant.Recently,a notice issued by China Central Television revealed that excessive chromium contaminated a list of common used drug capsules.Although the safety content standard for chromium has been set by Chinese Pharmacopoeia,it does not take in consideration the synergistic interaction of Cr with other chemical drug elements.In this research,we studied the mutagenic synergism of hexavalent chromium Cr (Ⅵ) with 1-(4-amino-2-methyl-5-pyrimidinyl) methyl-3-(2-chloro-ethyl)-3-nitrosourea (abbr.as ACNU),a kind of nitroso drug used to treat malignant gliomas,using Salmonella typhimurium tester strains TA 98 and TA 100.The results showed that Cr (Ⅵ) and ACNU were found to be mutagenic for TA 98 and TA 100 strains without metabolic activity.In the separate test,the minimal mutagenic dose of Cr (Ⅵ) and ACNU for TA 98 was 5 μg per plate and 50 μg per plate,and for TA 100,was 2 μg per plate and 10 μg per plate,respectively.In the combined experiments,the cooperative mutagenic dose of Cr (Ⅵ) and ACNU for TA 100 was down to 1 μg per plate.And for TA 98,Cr (Ⅵ) at the dose of 2 μg per plate combined with ACNU at the dose of 5 lμg per plate could induce significant genotoxic activity.It was observed that the revertant mutation effect of the combined treatment with low dosages of Cr (Ⅵ) and ACNU surpassed by far the mutation effect in the individual test.Based on our results,we suggest that the potential hazard for chemicals used in medical therapy should be reevaluated and their synergetic interactions taken in consideration.
