Lignin Biodegradation by Sphingobacterium.HY-H, Screening from Aerobic Sludge of Pulp and Paper Mill

  Six lignin degrading bacterial species,screening from activated sludge of pulp and paper mill,were isolated on mineral salt medium with lignin as the sole carbon and energy source.By comparing the results of the biodegradability tests,strain HY-H,identified as Sphingobacterium species by biochemical tests and 16SrRNA gene sequencing,was selected for subsequent tests on lignin biodegradation mechanism.The results showed that the strain HY-H was grow well under the condition:pH 7.0,temperature 30℃,and lignin/ammonium chloride ratio of 5.Under the optimum conditions,the maximum degradation capacity of the strain HY-H reached to 30%.GC-MS analysis indicated that after incubated with Sphingobacterium.HY-H,low molecular weight acids and lignin-related monomer compounds such as butyric acid,propylmalonic acid,acetovanillone were found,which strongly confirmed the degradation of lignin by Sphingobacterium.HY-H.
