Perchlorate-Induced Oxidative Stress in Isolated Liver Mitochondria Responsible for Apoptosis in Primary Cultured Hepatocytes

The Perchlorate (ClO4-) has been manifested that it could inhibit thyroid hormone synthesis. However, little is known about the effects of the Perchlorate on cell and organelle. To investigate whether Perchlorate exposure causes cytotoxicity in fish by inducing oxidative stress, the primary cultured hepatocytes isolated from Carassius auratus and were incubated with different concentrations (0-10 mmol L-1) of the Perchlorate at 25℃ for 12 h in vitro. In addition,to elucidate the precise mechanism of the Perchlorate induced oxidative stress in mitochondria responsible for toxicity in hepatocytes, isolated liver mitochondria were incubated with different concentrations (0-1000 μmol L-1) of the Perchlorate.
