Study on the Effect of Copper Stress on MLH1, MSH2 Promoter Methylation of Arabidopsis Thaliana

  This paper adopts improved bisulfite DNA sequencing technique,combining with the morphological and physiological indices ofArabidopsis thaliana (Arabidopsis) seedlings,and studied the effects of Copper (Cu) stress on methylation of MLH1 and MSH2 promoters of mismatch repair genes of Arabidopsis thaliana seedlings,aimed to choose the biomarkers sensitive to Cu stress.The results showed that under Cu (0、0.5、3.0、5.0 mg·L-1)stress for 21 days,the root elongation growth was inhibited,but no significant changes were observed in leaf number and fresh weight of aerial parts.The total soluble protein content in shoots was decreased with increasing Cu concentration.The contents of chlorophylls increased at first and then decreased with the increasing Cu concentration,it displayed a negative correlation in total.
