Genotoxicity Study of Multi-walled Carbon Nanotubes in the Presence of Humic Acids

  Nanotechnology involves research,production and use of nanoparticles,nanofibers and nanotubes with at least one dimension less than 100 nm.In recent years,an increasing number of products made ofnanostructures have entered the market leading to increased concem about their environmental fate and possible effects on human health.The present study aims to examine the possible genotoxic effect of Multi-Walled Carbon Nanotubes (MWCNTs) in the presence of a well-characterized Humic Acid (HA) isolated from Greek Lignites.The possible genotoxic effects of MWCNTs,HA and their mixture were studied in cultured human lymphocytes applying the cytokinesis block micronucleous (CBMN) assay for the detection of micronuclei (MN) in the cytoplasm of interphase cells.MWCNTs,HA and their mixture were studied at three different concentrations (5,15,25 μg/mL),(20,60,100 μg/mL) and (5+20,15+60,25+100 μg/mL) respectively.The fmdings of our study reveal that (a) MWCNTs and HA,each separately,were able to induce a minor increase in MN frequencies at all tested concentrations,compared with the control,(b) importantly,in the case of mixed MWCNTs and HA our results showed a three-to five-fold increase in MN frequencies compared with the control.
