Adventitious Shoot Organogenesis and Plant Regeneration by Immature Zygotic Embryos of Osmanthusfragrans

  Sweet osmanthus (Osmanthus fragrans Lout.), an evergreen shrub of Oleaceae family, is well known as an ornamental and economic plant for unique fragrance.The establishment of an efficient regeneration system and transformation system in sweet osmanthus for genetic improvement has not yet been reported.In this study, we firstly evaluated the effect of growth regulators on plant regeneration via organogenesis of sweet osmanthus.Immature zygotic embryos at cotyledonary stage were cultured on media with different concentrations of auxins and cytokinins.The results showed that the highest frequency of adventitious shoot regeneration (30%) was found in the 3.0 mg/L 6-BA+0.5 mg/L TDZ+0.5 mg/L NAA after 6 weeks dark treatment, and then shoots grew and prolonged in MS medium after another 6 weeks under light.The regenerates rooted when in the medium containing MS+2.0 mg/L NAA or MS basal medium and developed successfully after two months.
