Research on Construction of Tissue-engineering Heart Valve Scaffold with New Detergent

  The methods and effect of construction of tissue-engineering heart valve scaffold with detergents are different,and the method of detergent combining with the nuclease is more suitable than others.Objective of this study was to investigate the impact of different decellularization protocols on extracellular matrix integrity of xenogeneic tissue.Fresh porcine pericardium tissues were treated by sodium deoxycholate in combination with Triton X-100(SDT),sodium dodecylsulfate (SDS),and Octyl-glucopyranoside(OGP).After a subsequent nuclease digestion,the samples were dipped in phosphate buffered solution(PBS) to remove cell-debris.HE staining was used to detect whether the cells removed completely,Weigert staining was used to evaluate damage level of collagen fiber and elastic fiber,DNA content was examined by method of DNA extraction.Mechanical properties and biochemical compositions of extracellular matrix were also studied.SDS and OGP completely removed the cells,however,the effect of OGP on collagen fiber and elastic fiber was light,and SDT inadequately removed the cells.This suggested that the environmentally-friendly and non-toxic method of OGP could achieve both complete removal cells and preservation of the matrix structure,it might be a suitable approach to construct tissue-engineering heart valve scaffold.
