Extraction, Partial Characterization and Electorspinning of Acid-soluble Collagen and Pepsin-soluble Collagen From Mylopharyngodon Piceus Swim Bladder

Black Carp (Mylopharyngodon piceus) is one of the main species of freshwater fish produced in China.Acid-soluble collagens (ASC) and pepsin-soluble collagen (PSC) were prepared from the swim bladder of the black carp. The yield of ASC and PSC were 6.9% and 12.5% (wet weight basis),respectively.According to the SDS-PAGE electrophoretic pattern and H-NMR,both collagens contained α-and β-chains as their major components, and were characterized as type Ⅰ collagen with the cross-link of two α-chain.Collagen dissolved in hexafluoroisopropanol (HFIP) were electrospun. Its found that diameter of fibers is related to the addition of the collagen, positive voltage, the distance between aluminum foil and needle,as well as the flow rate of the solution.Because collagen can stop bleeding quickly,so the fibers can be used as medical hemostatic materials. The results also suggest that the swim bladder of black carp collagens have potential as an alternative source of collagen for use in various fields.
