Protein Extraction Methods Suitable for Two-dimensional Gel Electrophoresis Analysis on Protein Expression in Tomato Pedicel
In order to develop a suitable and effective protocol of protein extraction for proteome analysis on protein expression in tomato pedicel,six protocols for total protein extraction in tomato pedicel were compared,including TCA acetone precipitation method,Tris-HCL method,phenol method,Trizol precipitation method,Tris-acetone-phenol method and Urea-thiourea method.The extracts were separated by two-dimensional gel electrophoresis followed by Commassie blue staining.The results indicated that the highest protein yield was obtained by Trizol precipitation method,which significantly improved protein content in extract towards to TCA acetone precipitation method,but the other 4 methods gained lower protein yield at various levels.As far as spots detected in Commassie blue staining gels were concerned,Trizol precipitation method also performed well in the six methods.Moreover,with Trizol precipitation method,a stable,clear and repeatable protein spot could be gotten after Commassie blue staining with slightest tailing phenomena.So,Trizol precipitation method is the most suitable method for total protein extraction of tomato pedicel.
