Plant Regeneration from Leaf Explants of Sophora japonica ‘Liao Hong’

  A protocol for plant regeneration from leaf explants of ‘Liao Hong was developed.Most shoots directly regenerated at the base,edges and cut edges of leaf segments.The maximum shoot induction frequency (86.1±4.8%) and the largest number of shoots per explant (28.5±3.7) were obtained on Murashige and Skoog (MS) medium supplemented with 3.0 mg l-1 6-BA and 0.1 mg l-1 IAA.The largest number of new microshoots (4.6±0.59) was produced on MS medium supplemented with 1.0 mg 1-1 6-BA and 0.1 mg 1-1 NAA for shoot multiplication.The highest rooting rate (46.7±5.5%) of in vitro shoots was recorded on half-strength MS medium supplemented with 0.3 mg 1-1 IBA.The rooting induction ability was inhibited by addition of activated charcoal in the rooting medium.After transplanting,86% of plantlets survived in a mixture of 1:1 (v/v) vermiculite and peat moss.Although further study on improvement of the rooting rate remains,this is the first report so far for a successful plant regeneration of S.japonica ‘Liao Hong.
