A SCAR Marker for Detection Resistant Gene Ty-3 in Tomato
Tomato yellow leaf curl disease (TYLCD) is a new disease and causes important yield losses in tomato (Lycopersion esculentum Mill.) in China.For breeding resistant hybrids,PCR-based markers often were used for selection resistant materials.Most of the commercial cultivars resistant to TYLCD carry Ty-3 gene.In order to obtain molecular marker of resistant gene Ty-3,3 homozygous resistant lines (Ty-3/Ty-3) and 3 homozygous susceptible lines (ty-3/ty-3) were used for PCR reaction.The result showed that homozygous resistant lines produced about 600bp fragment,and homozygous susceptible lines produced about 320bp fragment.So this marker could distinguish resistant and susceptible lines,and was a co-dominant marker tightly linked to Ty-3 gene.The marker was used to detect 24 important inbred lines and the result showed that all of them did not carry Ty-3 resistant gene.23 individuals of F2 segregation population from resistant hybrid were also be detected with this marker,and the result showed that 16 individuals carried Ty-3 resistant gene,including 4 homozygous resistant individuals.Its concluded that this marker is a useful SCAR marker for detecting Ty-3 resistant gene in tomato.
