Establishment of Regeneration System from Inflorescence Explants of Hippeastrum Hybridum ‘Red Lion in vitro

This study examined the suitable combination of 1-naphthaleneacetic acid (NAA) and 6-benzyladenine (BA) and the effect of culture condition in vitro propagation ofHippeastrum hybridum cv.‘Red lion.Four different combinations of NAA (2.0,4.0 mg/l) and BA (2.0,4.0 mg/l) as well as two culture conditions (light and darkness) were tested. This paper found two ways to accomplish the regeneration from inflorescence explants in vitro:direct bulblet or plantlet induction, callus to adventitious bud formation. The petals showed a higher initiation ratio of adventitious shoot on MS supplemented with 2.0 mg/l NAA and 4.0 mg/l BA.4.0 mg/l NAA and 4.0 mg/l BA is conductive to callus induction on scape explants.Darkness is superior to light condition for initiation of petal explants obviously and could induce higher number of adventitious shoots directly on scape.
