A Method of Mitochondrial DNA Isolation in Peach
Mitochondrial DNA is widely used in the studying field of mechanism of cytoplasmic male sterility,heredity breeding of plant.Based on the former researches and field experiences of breeding researchers,the extrachromosomal inheritance has shown remarkable influence on peach breeding.In order to study on germplasm identification and germplasm resources research via peach extrachromosomal inheritance,a repmducible,simple and convenient mitoehondrial DNA isolation method is expected.This study investigated exacting mitochondria and exacting and purifying mtDNA from fresh leaves of peach,respectively.The result indicated that after the high speed tissue machine,high purity mitochondria could be exacted by differential centrifugation and sucrose density gradient centrifugation; finally,the process is preliminarily obtained,which can satisfy normal experiment requirements.
