Selection of Reference Gene for Real-time Fluorescence Quantitative PCR in Dahlia Petals

  In order to accurately and quantitatively analyze expression characteristics of senescence-associated genes in dahlia petal,the expression stability of three reference genes (18S rRNA,β-actin and GAPDH) was studied through real-time fluorescence quantitative PCR (qRT-PCR) under the different growth phases of dahlia petal,including squaring period,early florescence,full-bloom stage and flower senescence stage.The results show that the RT-PCR products are entirely consistent with the expected size.GeNorm and NormFinder analyses reveal that β-actin is the relatively more stable reference gene,and can be used for qRT-PCR of DpXTH gene expression related to petal senescence in dahlia.
