Optimization of Two-dimensional Electrophoresis for Proteome of Cucumber Cotyledon
2-DE analysis of complex plant proteomes has limited dynamic resolution because only abundant proteins can be detected.In order to improve the detection rate of cucumber cotyledon low-abundance proteins for 2-DE,we report two different fractionation protocols for preparation of whole-cell proteins,differential polyethylene glycol (PEG) precipitation method and differential urea concentration method.After 2-DE and PDQuest analysis,one time more protein spots can be detected after each fraction method and Rubisco is enriched in one ffraction.The extraction and fractionation techniques are simple and versatile and can be used to visualize more proteins in 2-DE analysis of plant proteome.Our studies suggest successful depletion of Rubisco can be achieved using a similar fractionation protocol.
