Characterization of a Novel Glycerol-3-Phosphate Acyltransferase from Lilium dauricum and Evolution Analysis with Those from Other Plants
The glycerol-3-phosphate acyltransferase genes (GATPs) are a kind of genes related with cold resistance of plants, which can be expressed by cold stress.A new Li-GAPT gene was cloned from Lilium dauricum in this research.Using Leaves as materials,a glycerol-3-Phosphate (GPAT) gene was cloned from the tissue culture regeneration plantlets of Lilium dauricum by the method of RT-PCR and rapid amplification of cDNA ends, and sequence analysis and homology comparison of the cDNA were performed by using DNArnan software and BLASTN program. The cloned full-length cDNA was 1544bp with 5-UTR of 10bp,an open reading frame of 1233bp,3-UTR of 31l bp and poly (A) tail of 19bp.The cDNA encoded 410 amino acids, and had a molecular size of 46 KDa.Its predicted that the differences between the 43 and 359 amino acid residues had a relationship with the differences of cold-stress in Lilium. The nucleic acid sequence analysis showed that it shared high homology with other known GPATs.
