Optimization of AFLP System for Tomatoes Based on Orthogonal Design

  Two inbred lines of tomato (Lycopersicon esculentum Mill.) FL1 and MFK1 with obviously different contents of lycopene were used in this experiment.In order to explore the amount of DNA,digested and connected time and pre-amplification product of dilution times,optimization of AFLP markers system were analyzed on the amplification results of the AFLP system The results showed that digested and connected time and the amount of DNA have the greatest impact on the amplification of the AFLP markers,and digested and connected time is the most critical factor for the PCR amplification results of this test.AFLP reaction system is established for tomatoes:300 ng of DNA template,6 h of digested time,pre-amplification products be diluted 40-fold.Bands amplified were stable and clear,so which can be applied to the tomato genetic map construction and tomato genetic diversity analysis.
