Identification of conformational IgG-binding epitopes on buffalo beta-lactoglobulin by web tool of MIMOX
Currently,there is no detailed information on the conformational epitope on β-lactoglobulin,and buffalo milk safety was highlighted with its increasing consuming.This study focuses on the generation of mimotope of buffalo β-lactoglobulin to characterize the conformational epitope by the biopanning of phage display peptide libraries combined with a molecular modeling approach.A total of 31 mimotopes on buffalo β-lactoglobulin were produced from the biopanning of a disulfide constrained heptapeptide phage display library,and two of them were predicted to be conformational epitopes candidates on the basis of bioinformation analysis by the use of DNAStar and web tool MIMOX,followed by be defmed into 7 conformational epitopes on buffalo conformational epitopes.The conformational IgG-binding epitopes were located in the two regions on buffalo beta-lactoglobulin and five amino acid residues PL-ENK in the comformational epitopes were identified to be conserved.We also found that two conformational epitope sequences were composed of five hydrophilic amino acids and two hydrophobic amino acids,which were corresponding to polar and non-polar,respectively.Our work suggested that some amino acids were more prone to be the part of conformational epitope,such as Proline,Leucine,Asparagine,Glutanube and Lysine.
