Microbial Total DNA Extraction and Purification from Aerobic Compost
Total microbes DNA were extracted from vegetable waste compost and purified by spin-bind cartridges.The modified DNA extraction yielded more than 76±5.8 (μg crude DNA/g compost) and 61±4.5 (μg purified DNA/g compost),more than 98%humic acids were efficiently removed by the purification.The gel electrophoresis of purified DNA had a length of about 23kb,and the band intensity roughly reflected the changes of DNA genome content in compost processing.The amplification of 16S rDNA V3 region showed the purified DNA was sufficiently free of contaminants to allow PCR amplification.These results indicated that the modified method to extract and purify compost genomic DNA were efficient and could be used for molecular analysis of complex microbial communities.
