Optimization of Extraction Procedures for EPS Extraction of Desulfovibrio Desulfurican
The efficiencies of extracting extracellular polymeric substances (EPS) from Desulfovibrio desulfurican was compared by using ethylene diamine tetraacetic acid disodium salt (EDTA),cation exchange resin (CER),NaOH,H2SO4,heating and sonication.Results showed that heating was most effective in extracting EPS.The carbohydrate,protein and nucleic acid in the EPS solution were 4.83,12.62 and 4.80mg/g dry cells,respectively.Results indicated that the EPS content were greatly dependent on the extraction method.UV-visible spectrum of the EPS extraction suggested that few cells were destroyed and lysis did not occur.UV-visible spectrum analysis,an easy and rapid technique,can be used to monitor cell lysis during EPS extraction from D.desulfurican
