Autophagy and Apoptosis of Alveolar Macrophages in the Different Stage of Silicosis

Objectives Autophagy activity changes and apoptosis in alveolar macrophages (AM) derived from the patients of different stage of silicosis were studied.Methods Thirty silicotic bservers and 30 silicosis patients at stage I , II , El were chosen as the research subjects.The information of occupation history was surveyed, and AMs of all subjects obtained from the massive whole lung lavage fluid were purified and cultured for 24 hours in vitro.After that the morphologies of autophagy precursor, autophagy body, autophagy - lysosome were observed under transmission electron microscope, early apoptosis of AMs was detected by using of Annexin V - FITC/PI kit.Results Autophagy body around the nuclear in AMs of observers was observed. Autophagy precursor in AMs of silicosis patients at stage I was observed, but other cytoplasm organelles was rarely be seen.Autophagy body and autophagy - lysosome in silicosis at stage II was also observed, different size of primary lysosome could be seen in patients with silicosis at stage HI, but typical autophagy precursor, autophagy body and autophagy — lysosome wasnt observed the.The AMs apoptosis rate in observers, silicosis patients at stage I , II , III were 0.20% ~ 15.28% , 0.58% ~ 18.13% , 1.07% - 19.56% , 2.52% -23.92% , respectively, and differences had statistically significant ( P < 0. 01).Conclusions There were difference of autophagic activity and apoptosis in Ams from different stage of silicosis patients, AMs autophagy and apoptosis may play an important role in the development of the silicosis.
