Establishing a Core Collection of Pleurotus ostreatus Cultivated in China Based on SSR Markers

To improve the utilization efficiency of Pleurotus ostreatus germplasm, a method for constructing the core collection of P. ostreatus based on molecular marker data was proposed. Using genotype information at 11 SSR makers for 48 P. ostreatus cultivars, an allele preferred sampling strategy was used to construct a P. ostreatus core collection using the UPGMA cluster method according to the genetic distances and the phenotypic traits. The size of each core collection was determined when the ratio of SSR alleles retained was maintained at 100% , 95% , 90% , 85% and 80% respectively. The ratio of SSR rare allele retained and the t-test results of Neis gene diversity and Shannons information index were used to evaluate the representativeness of the core collections. This was followed by analyses of the phenotypic representativeness of the core collections, including the coincidence rates of range, mean, and standard deviation between the core collections and the original samples. Our analyses identified that the allele preferred sampling strategy could be used to construct the most representative core collections with the least samples when the ratio of SSR alleles retained was maintained at 95%. In brief, the results indicate the following strategy as a suitable method for constructing P. ostreatus core collection: using allele preferred sampling strategy to sample, determining the size of core collection according to ratio of SSR alleles retained, finally validating representative of the core collection with some phenotypic evaluating parameters.
