Molecular Cloning and Sequence Analysis of DNA Topois-omerase II Gene from Omphalotus nidiformis and Its Application to Phylogenetic Analysis

A fragment of TopoⅡ was amplified from an Omphalotus nidiformis cDNA clone by nested polymerase chain reaction and primer walking. The full-length gene sequence was obtained with 3 and 5-full RACE-PCR. The length of open reading frame is 4 674 bp. The analysis of nucleotide acids sequence exhibits strong sequence homology to other eukaryotic topoisomerase II, and shares 44% -65% sequence consistent and 60% -78% sequence similarity with the reported TopoII. It shares 65% sequence identity with TopoII from Pleurotus ostreatus (ABG73430) and 62% identity with the one from Coprinus cinereus (BAD86854) at the amino acid level. Topo II from P. ostreatus and 0. nidiformis have the same signal sequence with eukaryotic Topo II (LIVMA-X-E-G-DN-S-A-X-STAG) , also have Toprim region and tyrosine activated site.
