Characterization of Mating-Type Genes,Determination of Nuclear Phase and Strain Identification in Rhizopogon roseolus

In the current studies, we sequenced and characterized the genes for homeodomain protein (RrA1-hox1, RrA1-hox2) and its flanking region mip gene in a bipolar ectomycorrhizal basidiomycete, Rhizopogon roseolus. Linkage analysis indicated that the sequenced region is mapped to A mating type locus in R. roseolus. By nucleotide alignment analysis of partial nucleotide sequences of two Rrhox1 genes from two mating compatible monokaryotic strains, we deduced 3-region of Rrhox1 gene sequences in R. roseolus has a divergence-homogenization duality, which makes the restriction fragment length polymorphism ( RFLP) analysis of this region possible. Using the primer pair designed according to 3-region of RrAl-hox1 and mip gene in A1 mating type, all ten dikaryotic strains of R. roseolus collected from nine prefectures of Japan were got the target PCR amplification products. After digesting the target PCR products by two restriction enzymes, AluI and RsaI, respectively, both RFLP patterns showed diversity and ten R. roseolus strains were well distinguished. So we concluded that 3-region of homeodomain protein gene ( Rrhoxl ) and mip gene is suitable for strain identification by RFLP analysis in R. roseolus.
