Lignosus rhinocerus (Cooke) Ryvarden-A New Potential Medicinal Mushroom that Can Stimulate Neurite Outgrowth at Low Concentrations of Extracts

Alzheimers disease is characterized by the loss of neurons and synapses in the cerebral cortex and some subcortical regions. Research has shown that absence of nerve growth factor causes Alzheimer-like symptoms in the adult brain of mice. A well known medicinal mushroom, Hericium erinaceus was found to have active compound/s with NGF-like properties and able to cross blood-brain barrier. The aim of this study was to investigate a new potential mushroom that had been used for medicinal purposes by local community in Malaysia. Neurite outgrowth in cultured rat pheochromocytoma cells, PC-12 acts as an indication of regenerative potential. Differentiated cells with one thin extension at least double the length of the cell diameter were scored positive. Our results showed that maximum outgrowth was shown at 20 μg/mL (w/v) for both L. rhinocerus aqueous and ethanol extract whereas for H. erinaceus maximal outgrowth was shown at 50 (μg/mL (w/v) of aqueous extract and 70 μg/mL (w/v) of ethanol extract. Lignosus rhinocerus extracts showed maximal neurite outgrowth at lower concentrations compared to H. erinaceus extracts. This showed that L. rhinocerus extracts may contain compound/s that able to enhance neurite outgrowth activity at a lower concentration. MTT assay was performed to check the percentage of cell viability after treatment with L. rhinocerus extract. IC50 value of aqueous extract of L. rhinocerus was recorded at 445. 15 μg/mL (w/v) and R value was 0. 988 4 while IC50 value of ethanol extract of L. rhinocerus was 282. 15 μg/mL (w/v) and R value was 0.991 8. IC50 values showed that extracts tested had no cytotoxic effect at low concentrations. To our knowledge, limited work has been done on neurite outgrowth activity using L. rhinocerus. Based on this preliminary result, further development and investigations are needed to confirm the mechanism / pathway for the neurite outgrowth. To our knowledge, this is the first report on neurite outgrowth stimulation activity by Lignosus rhinocerus.
