Quantification of DPPH Radical Scavenging Activity, Total Phenolic and Protein Contents of Cultivated Pink Oyster Mushroom (Pleurotus salmoneostramineus) on Different Rice Husk Percentages

This project aims to investigate the effect of different rice husk supplementations in the mushroom substrate bags towards the harvesting time, antioxidant, phytochemical and protein content of each flush of Pleurotus salmoneostramineus. In this study, the mycelium was obtained by tissue culturing a fresh Pleurotus salmoneostramineus. The mycelia were then inoculated into different substrate bags containing sawdust with different percentages of rice husk as supplement. The biological efficiency, antioxidant activities, phytochemical screening and protein content tests were conducted. Protein content determination was according to Kjeldahl Method with the conversion factor of 4. 38. In this experiment, it was found that using 20% rice husk as supplement resulted in the highest mean fresh weight at 110.91 g, highest mean biological efficiency at 45. 12% as well as the shortest growth time of about 68 days making it the most economical rice husk variable to be used commercially. Phytochemical screening revealed the presence of saponins. On the other hand, using 35% rice husk as supplement resulted in highest mean total phenolic content of 65.41 mg GAE/ g of sample extract as well as the highest mean DPPH scavenging percentage at 71. 58%. Highest protein content at an average protein per flush of 20. 29 ±5. 84% was recorded by Pleurotus salmoneostramineus grown using 35% rice husk as supplement as well. Correlation studies indicated that there was a strong positive correlation between percentages of rice husk used as supplement with protein content and antioxidant activities.
