The Effects of NAA and BA on the Induction and Differentiation of the Buds of Taxus Cuspidata
Young shoot tips and stem segments with buds of newborn Taxus cuspidata were regarded as explants. Growth hormone NAA and Cytokinin BA of different concentrations were added to B5 medium which was taken as basic medium in this study. Then single factor experiments were conducted. The results showed that: for callus induction the best concentration of NAA is 2mg/L and the induction rate was as high as 95.52%, however, the effects were not obvious for bud germination when using NAA separately. BA had a significant role in promoting for axillary bud germination. High concentrations of BA inhibited the growth of the buds. Germination rate of axillary bud of stems with buds increased significantly as high as 95.00% when using BA(4mg/L) and NAA(2mg/L) mixed. But browning phenomenon was serious. Then it should be transferred to the subculture after seven days.
