Development and Experimental Research of the Bioreactor Using Taraxacum Kok-saghyz Laticifers
Taraxacum koksaghyz Rodin (Russian dandelion) is one of the most important major rubber -producing plants in the world. Its laticifers can be act as bioreactor to study the overexpression or down regulation of key genes that are involved in rubber biosynthesis. As a model rubber plant, the experiment can be designed in two steps. One step is to construct the tissue culture system of T. koksaghyz, the other step is to construct the genetic transformation system and then transform plant expression vector into the T. koksaghyz. In this paper, plant expression vector pBI121-T14 containing the 35S promoter was selected to transfer into T. koksaghyz. The results showed that the tissue culture and the genetic transformation system of T. Koksaghyz have been successfully established, and the pBI121-T14 containing the 35S promoter was transformed into T. Koksaghyz by leaf disc transformation method. The expression product of GUS gene has been detected in transgenic T. Koksaghyz. In the following research, the plant expression vector (including Taxol synthase gene) with latex-specific promoter will be transferred into the T. Koksaghyz to further study about the mechanism of bioreactor.
