Measurement of Intracellular Nitrate Concentration in Cucumber Leaf using Nitrate-selective Microelectrodes
The manufacture and use of double-barreled microelectrodes, which are capable of simultaneous in vivo measurement of intracellular activities of NO3- and cell membrane potential (Em) of cucumber leaf, were described. Scanning electron micrographs showed that the electrode tip was not easy to be destroyed by the epidermis hair of cucumber. When filled with 100 mM KCI, all three barrels simultaneously reported identical transmembrane potentials, showing that all two tips were located in the same subcellular compartment. Intracellular estimates of Em in cucumber epidermal cells obtained with these doublebarreled microelectrodes were indistinguishable from those obtained using single-barreled microelectrodes. Measurements made with double-barreled NO3-selective microelectrodes in epidermis cells of 30-d-old cucumber leaves grown in a nutrient solution containing 0.05mM K+ having mean NO3- activity values of 153mM. The results in this paper confirmed that the ion-selective microelectrode previously used to barley leaf can be used successfully in cucumber leaf. So the cucumber nutrition can be diagnosed using microelectrode technique.
