Studies on the Isolation, Purification of Cellulase Produced by Aspergillus Niger Ⅲ and Trichoderca Viride and their Enzymatic Properties
Isolation and purification of the crude cellulase produced by apsergillus niger Ⅲ and trichoderma viride Ⅰ, and the enzymatic properties were studied in the paper. The results: By means of ammonium sulfate salting-out methods and sephadex G-100 column chromatography, five elution peaks were obtained, namely peak 1~5. The peak 2 included endoglucanase and exo-glucanase. Peak3 included endoglucanase, exoglucanase, and B-glycosidase. Peakl, 4 were not active, peak 5 included endoglucanase and Bglycosidase. By DEAE FF weak anion exchange chromatography, a kind of endoglucanase was purified from peak 2, and its molecular weight was 61.5kO by SDS-PAGE cataphoresis. A range of pH 5.0~6.0, the CMCase activity of peak 2 maintained 70%~80% of initial enzymatic activity, and the optimum pH was 5.0;higher enzyme activity appeared at 30~50℃, the optimal temperature was 50℃, and the enzyme activity rapidly decreased when temperature was higher than 60℃.
