Isolation and Analysis of Fertility-related Genes of Brassica Campestris L. ssp. Pekinensis

For studying of the molecular mechanism of genie multiple-allele inherited male-sterility in Brassica campestris L. ssp. Pekinensis, suppression subtractive hybridization (SSH) and cDNA-amplified fragment length polymorphism (cDNA-AFLP) were used to isolate differentially expressed genes between fertile and sterile buds of the two-type line AB01. 40 high quality sequences were generated (33 were upregulated in fertile buds and 7 were up-regulated in sterile buds). According to BLAST screening and functional annotation, these genes were with homology to known proteins, such as flowers/buds development proteins, metabolize-related proteins, cell structure proteins, cell growth/division proteins, secondary metabolic-related proteins and other proteins. Differential expression patterns between fertile and sterile buds were selected and verified using RT-PCR. Results indicated that there were five speciallyexpressed genes in fertile buds and others were upregulated expressed genes either in fertile or in sterile buds.
