The Performance of Replaceable Linear Polyacrylamide(PLA) Gel Electrophoresis of DNA Analyzer

We report the replaceable Linear polyacrylamide(LPA) gel was synthesized in house; the inner surface of capillary was coated with the same sieving medium (LPA). The DNA analyzer which was designed by authors group to detect the samples of capillary electrophoresis (CE) with laser induced fluorescence (LIF); The data collected with indirect methods establishes confidence in the electroosmotic mobility values obtained. Constant electroosmotic mobilitie of 2.5 × 10-8m2V-1s-1 was determined by tests. The bubbles in the LPA gel capillary electrophoresis were observed and recorded. It was discovered the bubbles can be reduced and affect CE little. A good linear CE current increase with applied field strength indicated the negligible Joule heating effects were present. This work will help to further optimize and improve high performance DNA separation in microfluidic applications.
