The effects of combination of Octreotide Acetate and Carcinoma-Eliminating Injection on expression of PAKi Protein in Hepal-6 Cell
Objectives To research the effect of different concentration Carcinoma-Eliminating and CarcinomaEliminating combinate Octreotide Acetate on hepatoma (Hepal-6) cell,as well as the pathogenesis. Methods Observe the cellular morphology with hematoxylin eosin(HE) stain; detect the cellular activity with colorimetric MTT assay; inspect the chang of the expression of PAKi protein with Reversed Transcript Polymerase Chain Reaction(RT-PCR). Results With the concentration increasing, the group of single Carcinoma-Eliminating and Carcinoma-Eliminating combinate Octreotide Acetate,the Hepal-6 cell rounding,cell volume lessen, the inhibition of proliferation become higher with the dose-effect relationship method (P<0.05 ), and compare to the modeling group and the group of 50mg mL-1 single Carcinoma- Eliminating, the PAKi protein significantly decrease in the group of 50mg-mL-1 Carcinoma-Eliminating combinate Octreotide Acetate (P <0.05) ; In contrast to the control group ,the effect of single Octreotide Acetate is not prominent on cytomorphology and proliferation ( P>0.05 ) .Concultions The group of Carcinoma-Eliminating combinate Octreotide Acetate significantly higher than the group of single Carcinoma-Eliminating in equal dose. And the concentration of 50mgmL Carcinoma-Eliminating combinate Octreotide Acetate is the best concentration of pharmacodynamic action. And palying its role have to do with reducing the expression of PAKi protein.
