COMPARATIVE MITOCHONDRIAL PAOTEOMIC ANALYSIS OF HEPATOCELLULAR CARCINOMA FROM PATIENTS
Objectives: Liver cancer, the most important malignant tumor in China, has been explored deeply by proteomics. Although a lot of liver cancer associated proteins has been found, mitochondrial protein marker related to liver cancer still remains ill defined. Methods: In the current study, we enrich and analyze mitochondrial sub-proteome of 20 liver cancer patients carcinomas and non-carcinomas tissues by two-dimensional electrophoresis(2-DE) coupled with MALDI-TOF/TOF. Results: The results showed that 10 proteins were down-regulated and 6 proteins upregulated in the carcinomas group compared with the non-carcinomas group. The altered patterns of three identified proteins were validated by western blot and immunohistochemsitry, and analyzed by STRING database. Furthermore, 10 kDa heat shock protein (Hspl0), Single-stranded DNA-binding protein(SSBPl), Peptidyl-prolyl cis-trans isomerase A(PPIA) identified in this study may be closely related to protein folding and translation. It shows that changes of mitochondrial DNA replication and protein folding in liver cancer is also worthy of our study, not only changes in the signal pathway. Conclusion: Collectively, these results suggest that specific mitochondrial proteins are uniquely susceptible to alterations in abundance and carry implications for the investigation of therapeutic and prognostic markers. Further studies focusing on these identified proteins will be used to predict treatment response and reverse apoptosis resistance.
