Upregulation of DNA polymerase iota (Pol ι) in esophageal squamous cell carcinoma

The transcriptional regulation of low-fidelity translesion DNA synthesis (TLS) polymerases in esophageal cancer was investigated. We found significant overexpression of mRNA levels for polymerase zeta (Pol ?), RAD18, polymerase iota (Pol i) and polymerase kappa (Pol k) in esophageal carcinomas. Pol t was further studied in greater detail. The promoter of POLI, encoding Pol x, was hypomethylated and its expression was not regulated by methylation. Spl and Oct-1 binding sites were identified in the POLI promoter. POLI transcription was correlated with that of Spl, and cancerous tissues showed significantly higher Spl (~4-fold) but not Oct-1 binding affinity, relative to nontumor samples. Taken together, our data presented the expression profile of TLS genes in esophageal cancer and elucidated the mechanism for POLI upregulation.
