Preparation and evaluation of nitrendipine nanocrystals
Purpose: To develop a nanocrystal formulation containing nitrendipine nanocrystals for oral delivery. Methods: High pressure homogenization process was employed to prepare the nanocrystals. Results: The prepared nanocrystals significantly improved the solubility and dissolution rate of nitrendipine. The in vivo testing demonstrated that the AUC0→24 of the nanocrystals was approximately 40-fold greater than that of coarse powder. Conclusion: Transformation of nitrendipine coarse powder into nanocrystals is an effective approach to increase the apparent solubility and dissolution rate and, in turn, to enhance the oral bioavailability of nitrendipine.
