C-di-GMP pathway in the biomining bacteria Acidithiobacillus thiooxidans

Acidithiobacillus thiooxidans is an acidophilic Gramnegative y-proteobacteria involved in the bioleaching of metal sulfides. It is capable of forming biofilms on mineral surfaces. In Acidithiobacillus sp. the molecular mechanisms involved in biofilm formation are still unknown. However, in many Gram-negative bacteria, the intracellular levels of the second messenger c-di-GMP control the regulation of biofilm formation. These levels depend on the balance between the enzymatic activities of diguanylate cyclases ( DGCs) ( synthesis) and phosphodiesterases ( PDEs) ( degradation) . The signal transduction is carried out by two kinds of receptors: five protein families (PilZ, PelD, FleQ, LapD and CLP) and two riboswitches (GEMM). Previously, functional c-di-GMP pathways have been characterized in A. ferrooxidans and A. caldus. Here, our main goal was to characterize the presence of a functional c-di-GMP pathway in A. thiooxidans. A bioinformatic analysis of the genomic sequence of A. thiooxidans ATCC 19377 predicted genes potentially encoding DGCs and PDEs, as well as effector proteins with PilZ domains. A sequence comparison showed that key amino acids of identified domains are conserved in most sequences. These results suggest that A. thiooxidans possesses a functional c-di-GMP pathway.
