13C NMR study on glucose metabolic pathways of genetic engineered chemolithotrophic Acidithiobacillus thiooxidans expressing phosphofructokinase
Phosphofructokinase is over expressed in the chemoautotrophic bacterium Acidithiobacillus thiooxidans Tt - 7 ( PSDK - 1). The metabolism pathways of 13 C enriched glucose in recombinant Acidithiobacillus thiooxidans Tt - 7 ( PSDK - 1) were investigated by using 13 C NMR. Glucose metabolism network was constructed to elucidate the distinctive carbon metabolism pathways in the gene engineered chemolithotrophic bacterium. The engineered strain is able to utilize EmbdenMeyerhof ( EM) pathway and the incomplete tricarboxylic acid ( TCA) cycle to convert glucose into a series of intermetabolites, for example, amino acids.
