Isolation and characterization of novel Sulfobacillus strains from a commercial bio-heap operation during moderate thermophilic temperature transition

The success of industrial bioleaching operations where temperature generation occurs, success is strongly dependant on the availability of the correct microbial speciation and consequently the transition during the temperature increase. Identification of the available species within the temperature range from 45 -55℃1 is limited. Members of the genus Sulfobacillus have been shown to thrive under harsh and constantly changing environments such as bioleaching. An extensive search of moderate thermophilic species, especially Sulfobacillus, was performed on leachate samples from a commercial chalcopyrite bioleaching heap that successfully transitioned into moderate thermophilic temperatures. A wide range of enrichments and physicochemical conditions were tested, including heat shock and micro-aerophilic conditions, to enrich for endospore-forming cultures. Three different species of the Sulfobacillus genus were isolated and characterized. One of these isolates corresponds to a novel specie of Sulfobacillus with no ferrous iron oxidizing capabilities and only 93% identity ( according to 16S rRNA gene) to the closest known cultivated relative; S. acidophilus. A fourth specie isolated corresponds to a thermophilic heterotrophe that is also a novel specie of the Acidimicrobiaceae family. The finding of these novel isolates shows a more diverse and extensive range of microorganisms that can thrive and establish themselves successfully in moderate to thermophilic heap bioleaching.
