Analysis of Low-temperature-induced ESTs in the Long-style Tomato Line T431 by Suppression Subtractive Hybridization
The long-style morphology of the mutant tomato line T431 is subject to temperature changes. To understand the molecular basis underlying this phenotype, we constructed a subtractive library for T431 under lowtemperature stress by the suppressive subtractive hybridization (SSH) approach. A total of 295 unique ESTs were obtained from 429 subtractive clones. Sequence similarity searches against the GenBank database using BLASTn and BLASTx algorithm revealed that 258 ESTs are homologous to known sequences, which correspond to 203 and 55 genes with known and unknown functions, respectively. No homologous sequences were found for the remaining 37 ESTs. which may represent new genes. Functional classification of the 203 known genes indicates that many of them are involved in primary metabolism, energy metabolism, disease!defense responses, and signal transduction, while some of them play roles in processes such as protein synthesis and processing, material transportation, transcriptional regulation, and development.
