Browning Inhibition and Regeneration of Red General Fuji Apple

Experiments were conducted with shoot tips and single node explants collected from field growing Red General Fuji apple ( Malus domestica ) trees to study the effects of MS or I QMS medium, pretreatment with low tempreature, ascorbic acid ( VC ) or activated charcoal ( AC ) on browning inhibition. The influences of different concentrations of 6-BA and NAA combinations on shoot proliferation, and 6-BA and IBA combinations on bud regeneration from leaf were also investigated. The results showed that serious browning occurred in this cultivar during the first 2 days in culture. Browning was significantly reduced by pretreatment with 4°Cfor 12 hplus 3.0 g/L AC, which was entirely inhibited by frequent subculture to fresh medium at 2-dayinterval. The optimal medium for shoot proliferation is MS +6-BAI.0 mg/L + NAAO.I mg/L. And the highest shoot regeneration frequency from leaf was 23.33% with 3.71 shoots per explant, using medium ofMS+ 6-BA 2.0 mg/L+ IBA 0.4 mg/L.
