Genetic Diversity and Population Structure in Amygdalus Ledebouriana Schlecht.Using Sequence-related Amplified Polymorphism Markers
The genetic diversity of 120 Amygdalus ledebouriana Schlecht. individuals from 5 populations in Xinjiang was investigated by using sequence-related amplified polymorphism (SRAP) markers. 137 bands were amplified by II informative and reliable primer combinations, of which 126 were polymorphic loci. At species level, percentage of polymorphic loci (PPL) were 91.97%, Neis gene diversity (h) equaled to 0.3424, Shannons Information index (1) was 0.5056; At population level, the average value of PPL was 62.92%; the mean value ofh amounted to 0.2612. and the general average of I was 0.3783. Genetic coefficient of differentiation (Gst) among populations based Nets gene diversity equaled to 0.2370. which showed 23.7% genetic variation existing among 5 populations. The mean value of Neis genetic identity equaled to 0.8802, estimate of gene flow from Gst (Nm) was 1.6094. which was able to be considered that gene flow was unblocked among 5 populations, and this result was uniform with that obtained by using Neis gene diversity. UPGMA dendrogram based Neis genetic distance showed that genetic distance among populations was not to have significant correlation with geographic distance. The conclusion shows that germ plasm protection and exploitation strategies of Amygdalus ledebouriana Schlecht. should be prior to preserve available population, especially, to strengthen the screening and reserve the mutation of BRJpopulation.
