Characterization of Phalaenopsis aphrodite Chloroplast Microsatellites

Chloroplast simple-sequence repeats (cpSSRs) were investigated in the Phalaenopsis aphrodite. 61 SSRs were identified. Among them there were 47 (78.3%) mono-, 8 (15.3%) di-. 4 (6.7%) tri-, I (1.7%) tetra-, I (1.7%) penta-, 1(1.7%) hex-nudeotide SSRs, mainly distributed in large single-copy regions (LSC). The SSR frequency was approximately one SSR/2.4kb on an average. The monomuceleotide repeats appeared to be the most abundant. Five cpSSR primer pairs were designed and tested. The result showed that development of genus-septic cpSSR primers based on sequence from the model species was perfectly feasible.
