Optimization on Tissue Culture Technology of Cymbidium Kanran Makino
Using Cymbidium Kanran Makino as material, the effect of medium and plant growth regulator on PLBs proliferation, differentiation and root induction was studied. The result showed that 6-benzyl aminopurine (6-BA) had the most significant effect on PLBs proliferation. Improved M S (IM MS)+0.6mgL~ NAA+ 2.0mgL 6-BA was the best medium for PLBs proliferation with the highest proliferation rate of 8.36. Medium had the most significant effect on shoots development, and the best medium was IM MS+ 0.6mgLNAA+0.5mgL~6-BA with the highest germination rate of 6.70; at the same concentration of plant growth regulator, the use of 1/2MS medium could promote shoots growth. Activated carbon (AC) had the most significant effect on root induction, and IM MS+ 50g-L~ AC+0. lingL 1 IBA+0.5mg-L~6-BA was the best medium for root induction; the highest induction rate was 100%.
