Effects of Phytohormone Conditions on Callus Formation and Floral Differentiation from Styles in Crocus Sativus L.
In order to apply plant tissue cultural techniques on producing officinal compositions of saffron, styles of Crocus sativus Linn, as explants, the effects of various differentiation on callus formation and floral differentiation from styles were studied on the MS basic media, which was supplemented with various hormones, such as 6-BA , NAA, 2,4-D and IBA at different ratios. The results showed that these styles first dedifferentiated to callus on the suitable medium in early period, which further differentiated into bud-like structures, petal-like structures and stigma-like structures. It was found that NAA is the most effective auxin, and the following is IBA and 2, 4-D in the culture of styles. The ratio of 6-BA/NAA plays an important role in floral types, high ratio of 6-BA/NAA is helpful for callus to differentiate the stigma-like structures, while low ratio stimulates the formation of petallike structures.
